The assay of urinary peptides using a biuret reagent.

چکیده
IT HAS BEEN POINTED OUT PREVIOUSLY that the method for the assay of urinary peptides using the biuret reagent is a much simpler procedure than others in current use (1). However, a disturbing difficulty with this method has been the lack of reproducibility of the biuret reagent (1). This resulted in sporadic errors approximately 10 per cent and in a feeling of uncertainty regarding the method itself. A study was made, therefore, of the reproducibility of this reagent. In addition, considerably more information on interfering substances was gathered, and a study was made of the stability of peptides in urine specimens. MATERIALS AND METHOD Reagents

Direct analysis for urinary protein with biuret reagent, with use of urine ultrafiltrate blanking: comparison with a manual biuret method involving trichloroacetic acid precipitation.
We describe a method for measuring urinary protein with a centrifugal analyzer. Biuret reagent is used, and blanking with an ultrafiltrate of urine eliminates interferences from the nonprotein, biuret-positive chromogens in urine. We compare results by this new method with those by a manual method in which trichloroacetic acid precipitation and biuret reagent are used. The new method shows good...
متن کاملUrinary peptides; method for assay.
THE USUAL METHOD of peptide estimation requires the assay of a protein-free filtrate for amino-acid concentration before and after hydrolysis. Peptide amino acid is then calculated by difference (1). In another method (2) specifically designed for urine, the peptides are precipitated with phosphotungstic acid from a trichloroacetic acid filtrate. The precipitate is dissolved and color is formed...
متن کاملCross-reactivity of amino acids and other compounds in the biuret reaction: interference with urinary peptide measurements.
BACKGROUND Biuret assays for total protein measurement are considered to react with all peptides longer than 2 residues. Some studies using biuret assays of urine suggest that small peptides generally are more abundant than proteins in urine, but it is not clear whether this is a problem of assay specificity. METHODS We analyzed the specificity and kinetics of a biuret reaction for solutions ...
متن کاملImproved biuret procedure for routine determination of urinary total proteins in clinical proteinuria.
This communication describes and evaluates an improved routine methodology for quantitating clinical proteinuria. Based on investigations of Piscator and of Savory et al., a modified Tsuchiya's reagent (ethanolic HCI-phosphotungstic acid) is used to precipitate proteins at 56 degrees C, followed by biuret spectrophotometry at 540 nm. The accuracy of the proposed procedure was assessed by compa...
متن کاملEstimation of peptide concentration by a modified BCA assay
Although biuret based protein assays are theoretically applicable to peptide measurement, there is a high level of inter-peptide variation, determined largely by peptide hydrophobicity. This variation in peptide reactivity can be significantly reduced by heat-denaturation of peptides at 95 °C for 5 minutes in the presence of 0.1 M NaOH containing 1% (w/v) SDS, prior to incubation for 30 min at ...
